Humanin side effects and risks

A peptide the body makes; injection benefits and safety remain unproven.

Also called: HN, MT-RNR2 peptide.

Mitochondrial-derived peptides · 24-amino-acid mitochondrial cell-survival peptide.

Reported side effects

  • Injection-related allergy or infection: General unapproved injectable risk; no humanin specific event rate., serious. Breathing difficulty needs emergency care; spreading painful redness needs urgent review. (source)

Stop and get help if

  • Trouble breathing, throat or tongue swelling, collapse: Stop exposure and call emergency services now. Use prescribed adrenaline if available.
  • Spreading hot, painful injection-site redness, especially with fever: Stop injections and seek same-day care; confusion or fainting needs emergency care.
  • Confusion, seizure, collapse or low blood sugar that does not resolve: Call emergency services for severe symptoms; follow the established diabetes rescue plan if applicable.

Who should avoid it

  • pregnant: Precaution: no administered-peptide pregnancy safety data were identified.
  • breastfeeding: Precaution: transfer into milk and effects on a nursing infant are unknown.
  • tryingToConceive: Precaution: human fertility and embryo safety after injected peptide are unknown.
  • under18: Precaution: no pediatric dosing or injection-safety data were identified.
  • diabetes: Glucose effects and interactions with diabetes medicines are untested in humans.
  • kidneyDisease: Human clearance and dose adjustment in kidney disease have not been established.
  • liverDisease: Human exposure and dose adjustment in liver disease have not been established.
  • heartCondition: Cardiac biomarker studies do not establish injection safety in people with heart disease.
  • surgeryPlanned: Tell the care team about use; perioperative drug interactions and safety are unknown.
  • activeCancer: Precaution: humanin promoted glioblastoma growth and blood-vessel formation in mice; it also blocks cell-death pathways.
  • cancerHistory: Discuss recurrence risk; cell-survival signaling and a tumor-growth signal make long-term cancer safety uncertain.
  • testedAthlete: Prohibited at all times under WADA 2026 S0 for non-approved pharmacological substances.

Interactions

  • Insulin and sulfonylureas: Possible added glucose lowering is theoretical. No human interaction study has measured the risk.
  • Metformin and other glucose-lowering medicines: Combination effects on glucose, appetite and exercise tolerance are untested for injected humanin.
  • Anticoagulants and antiplatelets: No compound-specific interaction is established. Review injection-related bleeding risk with the prescriber.
  • Other mitochondrial peptides: No human trial establishes the safety or benefit of stacking mitochondrial peptides. HNG is not native humanin.
  • Cancer treatment: Precaution: cell-survival signaling may affect cancer biology; a glioblastoma model showed tumor promotion. Human combination safety is unknown.
  • Growth hormone and IGF-1 therapies: GH/IGF-1 can alter endogenous humanin. The safety of adding humanin injections has not been tested.

Product quality risks

  • Unapproved injectable products can have too much, too little or none of the stated ingredient.
  • Possible contaminants include microbes, endotoxins, solvents, metals and particles; risk varies by batch.
  • No independent market-wide test of native MOTS-c or humanin products was identified.
