Epitalon: research
A peptide with telomerase effects in cells, but no proof that people live longer.
Also called: Epithalon, Epithalone, AEDG, Ala-Glu-Asp-Gly.
Khavinson bioregulator peptides · Synthetic pineal tetrapeptide studied in telomere and circadian models.
Four human reports cover about three participant groups. Two night-shift reports likely reuse participants; longer human life is unproven.
Studies
- Retinitis pigmentosa: local eye injections. cohort, n=162, 162 examined patients aged 18–72 with congenital retinitis pigmentosa. An old controlled report suggests visual changes but lacks modern trial safeguards. (source)
- Older adults: 10 mcg IM course. rct, n=50, Older adults aged 60–79; a young reference group was also studied. A tiny IM study changed melatonin levels without showing better sleep or longer life. (source)
- Night-shift workers: sublingual spray. rct, n=75, 75 women aged 40–50 assessed; 40 with low melatonin metabolite randomized. A small biomarker trial does not establish insomnia treatment. (source)
- Night-shift workers: companion gene report. rct, n=75, 75 female doctors/nurses aged 40–59, mainly working night shifts. A related report shows gene-expression changes after sublingual exposure. (source)
- Telomerase in human fibroblasts. in-vitro, Human fetal fibroblasts in culture. The central telomerase claim began as a cell-culture finding. (source)
- Cultured cells beyond division limit. in-vitro, Human somatic cells in culture. A culture result cannot establish human longevity. (source)
- Telomeres in normal and cancer cells. in-vitro, Normal human cells and 21NT/BT474 breast-cancer cell lines. Independent cell work raises safety questions as well as mechanistic interest. (source)
- Isolated rat pineal glands: null result. in-vitro, Pineal glands from young and old rats. A direct melatonin-secretion effect was not reproduced in this rat tissue experiment. (source)
- Hormone rhythms in old monkeys. animal, Female rhesus monkeys of differing ages. Primate hormone changes do not establish human sleep benefit. (source)
- Spontaneous tumors in female mice. animal, One-year-old female C3H/He mice. A mouse tumor signal cannot settle human cancer safety. (source)

These are doses reported in labels, trials or user reports, not a personal plan. Check the source behind each one.
Epitalon side effects and risks
Reported side effects
- Sleep disruption, activation or anxiety: Anecdotal reports; no denominator or confirmed causality., moderate. Stop and seek review if symptoms persist or worsen. (source)
- Immune reaction to an injection: FDA identifies potential risk; no reliable human incidence., serious (source)
- Injection-related infection: No Epitalon specific incidence available., serious (source)
- Long-term cancer effects, unresolved: Mechanistic concern, not an established human adverse event rate., serious (source)
Stop and get help if
- Trouble breathing, throat or tongue swelling, or collapse: Stop use and call emergency services now.
- Fever with spreading injection-site redness, confusion or fast breathing: Seek urgent emergency care for possible severe infection.
Who should avoid it
- pregnant: No human pregnancy safety data. Precautionary exclusion.
- breastfeeding: Milk transfer and infant effects are unknown. Precautionary exclusion.
- tryingToConceive: Fertility effects and a safe washout interval are unknown. Precautionary exclusion.
- under18: Child safety is unestablished. Small reports do not establish a safe pediatric dose.
- kidneyDisease: Human clearance and dose adjustment in kidney disease are unknown.
- liverDisease: Human clearance and dose adjustment in liver disease are unknown.
- psychiatricCondition: Sleep disruption or agitation may worsen existing symptoms. Human psychiatric safety is untested.
- seizureDisorder: Seizure safety and interactions with seizure medicines have not been established.
- other: Avoid re-exposure after a serious allergic reaction.
- testedAthlete: Treat as prohibited under WADA S0. This applies the class rule; neither peptide is named.
- bleedingDisorder: Needle injections can cause bleeding or a hematoma. This is a procedure risk.
- activeCancer: Precautionary exclusion: telomere-maintenance effects in cancer cells raise concern. Human cancer risk is unknown.
- cancerHistory: Discuss with the oncology team. No human study establishes recurrence risk or long-term cancer safety.
Interactions
- Anticoagulants during intramuscular injections: IM injections can cause bruising or hematoma. A peptide-specific drug interaction has not been studied.
- Insulin and sulfonylureas: No human interaction study or dose-adjustment rule was found. Do not infer a need to change diabetes medicines.
- Melatonin and sleep medicines: Effects on sleep timing may overlap. No human combination or drug-absorption study establishes an interaction or adjustment rule.
- Other experimental peptides: No controlled combination safety evidence found. Sleep disruption is reported; multiple drugs obscure the cause.
Product quality risks
- A 2015 analysis identified Epitalon in two illegal preparations. Identity alone did not establish correct dose, sterility or safety.
- Unapproved injections can contain the wrong drug, the wrong amount, microbes or bacterial toxins called endotoxin.
- A research-only label, clear solution or high purity percentage does not establish injection quality.
How Epitalon works
In cells, Epitalon can change telomerase and other ways cells maintain chromosome ends, called telomeres. Small human studies also found changes in melatonin-related markers. These findings do not show that people live longer or sleep better.
- Telomerase and ALT pathways: Telomere-maintenance changes seen in normal and cancer cell cultures.
- Melatonin and clock-gene markers: Small human studies show marker changes; clinical sleep effects remain unproven.
Half-life: No reliable human half-life or clearance study was found.
Gaps: No human lifespan trial of synthetic Epitalon was found. Long follow-up studies commonly cited used the different extract Epithalamin. Four exposure publications are listed. The two circadian reports likely overlap, giving about three independent cohorts. No ClinicalTrials.gov record or Cochrane review specific to synthetic Epitalon was located by the searched names. The eye study uses local parabulbar injections. This route cannot be represented as a protocol without mislabeling it in the current schema. The original eye paper says visual acuity rose 0.15–0.25. A 2025 review says 0.15–0.20; the primary figures are retained. The eye study’s examined, treated and eye-based denominators are not fully clear. Treated n and modern adverse-event rates are not inferred. The 2007 study initially examined 49 older adults; listed treatment groups account for 40. Disposition of the other nine is unclear. The 0.5 mg/day spray schedule is checked in FDA’s review and the companion primary report; original English Ivko full text was not acquired. No human SC dose or frequency ceiling was verified. Community SC protocols therefore have no invented studiedMax or aboveStudied comparison. Microdosing, five-on/two-off schedules, repeated high-dose cycles and lifelong use have no controlled human safety support. Human pharmacokinetics, absorption, fertility, pregnancy, breastfeeding and interactions are uncharacterized. Telomerase and ALT findings raise a theoretical cancer concern; they do not establish that Epitalon causes or prevents cancer in people. The 2025 cell paper has a November 2025 correction replacing Figures 1–3. Corrected findings still require independent replication. PCAC advised inclusion in July 2026 despite FDA staff opposition. No official minutes or final FDA listing found; exact vote counts are omitted. The orphan designation was withdrawn/revoked on January 6, 2016 per FDA’s 2026 briefing; it never conferred approval. No validated home reconstitution, product-specific mixed-vial storage time or routine monitoring schedule was found. No Epitalon-specific poison-center case series or representative contamination survey was found. Analytical identity in two preparations is not a quality survey. Community prevalence is not a measured population rate. Forum comments include corrected unit errors and unconfirmed planned increases. The 5 mg/day for 20 days and five-on/two-off reports omit a clear route. They are not converted into route-specific protocols. Safety blocks and warnings are precautionary screening judgments, not an approved label. Specific risks are identified as theoretical or unstudied. The 100 mcg/day for 10 days community schedule has no verified route in the cited threads, so it is not a route-specific protocol.
