How Tesofensine works

An oral appetite drug with weight-loss trial results and heart-rate and psychiatric risks.

Also called: NS2330, NS-2330, NS 2330.

Fat-loss drugs · Serotonin, norepinephrine and dopamine reuptake inhibitor.

It blocks the pumps that clear serotonin, norepinephrine and dopamine between nerve cells. This can reduce hunger and change mood, sleep and heart rate. It does not work through the GLP-1 pathway.

  • Serotonin transporter (SERT): Blocks serotonin reuptake.
  • Norepinephrine transporter (NET): Blocks norepinephrine reuptake; may increase heart rate and blood pressure.
  • Dopamine transporter (DAT): Blocks dopamine reuptake; human brain scans show transporter occupancy.

Half-life: About 234 hours in an Alzheimer’s population model; active metabolite M1 about 374 hours.
