An experimental cell-clearing peptide with mouse findings and no established human dose.

Also called: FOXO4 DRI, FOXO4-D-retro-inverso, FOX04-DRI, Proxofim.

Senolytic peptides · Experimental FOXO4-p53 interaction-disrupting peptide · Evidence: Animal studies only.

What FOXO4-DRI does

FOXO4-DRI is designed to kill some senescent cells, which are damaged cells that stop dividing. Mouse and cell studies report benefits, but no human dosing study was found. It has no approved dose or proven longevity benefit in people. Animal lung and liver harm signals make safety a major gap.

  • In mouse studies, it cleared some worn-out (senescent) cells and improved selected health measures. (animal)
  • In aged mice, it raised testosterone and improved sperm quality; this has not been tested in people. (animal)
  • People report trying it in hopes of clearing aging cells, but better health in people has not been shown. (Reported by users)

Uses in the sources

  • People self-experiment hoping to clear senescent cells. Longer life or better health in people has not been demonstrated. (Reported by users) (source)
  • Forum users seek joint or tissue benefits. Human cartilage-cell experiments did not show improved cartilage formation. (Reported by users) (source)
  • Some users track testosterone after use. Mouse findings do not prove higher testosterone or fertility in people. (Reported by users) (source)

FOXO4-DRI dosage

Typical reported dose: 3 mg every other day for six doses

  • Reported 3 mg on alternate days: 3 mg every other day for six doses every other day for six doses, under the skin. Reported by users, rare report. Studied in: Adult forum self-experimenters; one author describes use with a spouse. (source)
  • Reported 40 mg in one day: 5 mg, 5 mg, 10 mg and 20 mg on the same day four injections in one day; no repeat schedule stated, under the skin. Reported by users, rare report. Studied in: One adult LongeCity self-report. (source)

These are doses reported in labels, trials or user reports, not a personal plan. Check the source behind each one.

How to mix FOXO4-DRI

Comes as: Powder in vials or bottles, reported by self-experimenters..

Mixed with: Bacteriostatic water is reported in forums; no approved human diluent exists..

Common vial sizes: 10 mg. Common water amounts: 1 mL.

  • A 10 mg bottle appears in a Reddit report; this does not establish a standard vial size.
  • One report used 1 mL for 5, 10 or 20 mg portions. These are reported mixtures, not validated recipes.
  • The same report described pain and leakage. Acidic diluent was also painful.
  • Water, dilution or a clear solution cannot prove sterility or remove endotoxin.

Before mixing: No validated human-product shelf life or storage range found.

After mixing: No validated stability, sterility or discard interval after mixing was found.

FOXO4-DRI side effects and risks

Reported side effects

  • Injection pain and swelling: Forum reports only; no reliable rate or dose response estimate., moderate. Persistent or worsening pain, redness or swelling needs assessment. (source)
  • Low mood or disrupted sleep: One report after a 10 mg total course over three days., moderate. Stop exposure and seek care if symptoms persist or become severe. (source)
  • More frequent bowel movements: One person reported this for three days; no clinical rate., mild (source)
  • Possible harm to lung circulation: Mouse signal only; human incidence is unknown., serious. New breathlessness, chest pain or fainting needs urgent assessment. (source)
  • Possible liver injury: Repeated dose mouse preprint; human incidence is unknown., serious. A clinician may check liver tests after exposure; no validated testing schedule exists. (source)
  • Severe allergy or injection-related infection: General unregulated injection risks; no FOXO4 DRI specific rate., serious. Breathing problems need emergency care; hot, painful swelling needs same-day care. (source)

Stop and get help if

  • Swollen tongue or throat, trouble breathing, or collapse after an injection.: Stop exposure and call emergency services now.
  • Injection site becomes hot, painful or swollen, especially with fever.: Get same-day care. Call emergency services for confusion, fainting or fast breathing.
  • New severe breathlessness, chest pain or fainting.: Stop exposure and seek emergency assessment.

Who should avoid it

  • pregnant: Precaution: no pregnancy safety data or tested human dose.
  • breastfeeding: Precaution: passage into milk and effects on infants are unknown.
  • tryingToConceive: Precaution: mouse fertility findings do not establish safety around conception.
  • under18: No pediatric dosing or safety study. Newborn-rat experiments do not establish child safety.
  • activeCancer: Precaution: it changes p53 signaling. Human cancer-treatment effects are untested; involve the oncology team.
  • cancerHistory: Long-term effects on cancer surveillance and recurrence are unknown.
  • other: FOXO4-DRI worsened lung-vessel function in a susceptible mouse model; no human safety data.
  • heartCondition: Lung-circulation harm in mice makes heart or lung disease a reason for clinician review.
  • liverDisease: Repeated-dose liver toxicity appeared in a mouse preprint. No human dose adjustment is known.
  • kidneyDisease: Mouse kidney benefits do not establish safety or dose adjustment in kidney disease.
  • immunosuppressed: Unregulated injections can cause infection. Risks are higher with a weakened immune system.
  • surgeryPlanned: No perioperative safety data. Tell the surgical team about exposure and any injection-site reaction.
  • testedAthlete: Falls under the 2026 S0 rule for non-approved drugs; prohibited at all times.
  • other: Previous severe allergy to the peptide, diluent or injected product.

Interactions

  • Other senolytics: No human combination studies. Adding other cell-killing agents may add toxicity; this is a theoretical concern.
  • Cancer therapies: Mouse studies altered responses to chemotherapy or radiation. Human combinations and treatment timing are untested.
  • Anticoagulants: General injection-related bleeding concern, especially with muscle injections. No FOXO4-DRI drug interaction has been established.

Product quality risks

  • No independent peer-reviewed test-purchase study of online FOXO4-DRI products was found.
  • Online injectable peptides may have wrong ingredients, wrong amounts, solvents, particles, microbes or endotoxin.
  • Indirect evidence: three online semaglutide samples had 7.7%-14.37% purity and detectable endotoxin. These were not FOXO4-DRI.
  • An HPLC purity percentage alone does not prove identity, vial content, sterility or freedom from endotoxin.

What to expect

No human dosing study found. Studies of donated human cells or removed tissue are lab work, not human treatment studies.
