Studies
- A bolus resets LH timing. single-arm, n=13, 13 healthy adult men. One IV dose triggered an LH pulse and delayed the next spontaneous pulse. (source)
- First dose and infusion study. open-label, Overall bolus cohort: 6 healthy men; 4 men in each infusion experiment. IV dosing raised LH and testosterone; these experiments did not test home injection schedules. (source)
- Response depends on sex and cycle. pk-study, n=46, 11 healthy men and 35 healthy women; 4–5 per dose group. KP-10 cleared in minutes; women responded differently across the cycle. (source)
- Cycle phase changes the LH response. single-arm, Healthy women: early follicular n=10, preovulatory n=3, midluteal n=14; four additional higher-dose visits. Responses to the same dose varied by menstrual phase. (source)
- Contraception changes the response. open-label, n=24, 10 follicular-phase, 6 postmenopausal, 4 combined-pill and 4 progestogen-implant users. Combined-pill users had a blunted LH response. (source)
- NKB-deficient patients respond to infusion. open-label, n=4, 3 men and 1 woman with TAC3 or TACR3 loss-of-function variants. Continuous KP-10 restored LH pulses in four selected patients. (source)
- Low testosterone in type 2 diabetes. open-label, n=12, 5 men with type 2 diabetes and low testosterone; 7 healthy comparators; 4 men completed infusion. Hormones rose during an 11-hour infusion in four men with diabetes. (source)
- Most congenital hypogonadism did not respond. open-label, n=12, 11 adults with persistent congenital IHH; one man with reversal. An intact responsive GnRH network was needed for an LH response. (source)
- KP-10 versus KP-54 and GnRH. crossover-rct, n=10, 10 healthy men; 5 participants per dose group, with some crossover between doses. Both kisspeptins raised LH, but GnRH produced a larger response. (source)
- Reversal versus relapse of hypogonadism. case-series, n=6, 6 men with congenital IHH and prior spontaneous reversal. KP-10 response tracked whether natural LH pulses had returned. (source)
- Estrogen and NKB feedback. crossover-rct, n=20, 20 healthy women: 10 with NK3R blockade and 10 without. Estrogen and NKB signaling changed how long the LH rise lasted. (source)
- GnIH co-infusion in men. crossover-rct, n=4, 4 healthy men in the KP-10 experiment; 5 postmenopausal women had GnIH alone. GnIH did not reduce KP-10-induced LH release in four men. (source)
- 24-hour infusion after menopause. open-label, n=8, 8 postmenopausal women; 4 off estradiol, 7 on estradiol; 3 did both. Estradiol altered the response to sustained KP-10 exposure. (source)
- NKB blockade in men. open-label, n=6, 6 healthy men. KP-10 still stimulated LH despite NKB receptor blockade. (source)
- Infusion in two women with high prolactin. case-series, n=2, Two women with cabergoline-resistant microprolactinomas and chronic absent periods. A 12-hour infusion increased reproductive hormones in two women. (source)

- Delayed puberty: early response study. open-label, n=20, 15 children with delayed puberty; 5 healthy adult male comparators. Response varied widely in children with delayed puberty. (source)
- Response across puberty stages. open-label, n=30, 25 boys: 5 per Tanner stage; 5 adult men. The strongest LH response appeared late in puberty. (source)
- Sex steroids did not restore response. case-series, n=7, 6 men and 1 woman with congenital IHH. Sex-steroid replacement did not make congenital IHH responsive. (source)
- Aging and the testosterone response. open-label, Healthy adult, middle-aged and older men; registry enrollment 15. Older men had less testosterone response despite an LH rise. (source)
- Kisspeptin and opioid blockade in TAC3 loss. case-series, n=5, Five related women; four had TAC3 deficiency; three received KP-10 boluses. Kisspeptin responses persisted despite absent NKB signaling. (source)
- Irisin after a single dose. open-label, n=16, 8 normal-weight and 8 obese men. Irisin rose after dosing, but weight-loss effects were not tested. (source)
- Delayed puberty: later outcomes. cohort, n=16, 16 children previously tested for delayed puberty. The earlier LH response predicted pubertal progression in this small cohort. (source)
- PCOS: KP-10 with NKB blockade. crossover-rct, n=10, 10 unique women with PCOS; 8 had antagonist pretreatment and 7 had no pretreatment, with 5 women in both conditions. KP-10 still raised LH after NKB blockade in PCOS. (source)

Stop and get help if
- Trouble breathing, throat swelling, widespread hives or fainting after injection: Stop and seek emergency care now.
- Chest pain, sudden weakness, severe breathlessness or collapse: Seek emergency care now; do not wait for hormone tests.
- Severe muscle pain or weakness with dark urine: Seek urgent care for possible muscle breakdown.
- Fever with worsening redness, swelling or pus at an injection site: Stop injections and get prompt care for possible infection.
Who should avoid it
- pregnant: Precaution: pregnancy safety has not been established; KP-10 affects reproductive signaling.
- breastfeeding: Precaution: milk transfer and infant safety are unknown.
- tryingToConceive: Keep fertility use within a specialist plan; ovulation effects vary and a home conception schedule is unproven.
- under18: Research includes children, but no approved pediatric treatment or home dose exists.
- hormoneSensitiveCancer: Precaution: reproductive hormone stimulation may conflict with cancer care; no human cancer-safety data establish acceptability.
- activeCancer: Cancer effects vary by cell model. Human cancer safety is unknown; review any active cancer with the oncology team.
- heartCondition: KP-10 contracts isolated vessels and worsened plaque in susceptible mice. Human cardiovascular risk is unknown.
- uncontrolledBloodPressure: Vessel-contraction findings support caution; acute small trials cannot establish safety in uncontrolled hypertension.
- diabetes: A small diabetes study measured hormones, not diabetes outcomes. Glucose and medication effects need review.
- kidneyDisease: No validated kidney-function dose adjustment or repeated-dose safety data were found.
- liverDisease: No validated liver-function dose adjustment or repeated-dose safety data were found.
- testedAthlete: WADA 2026 S2.2.1 names kisspeptin under testosterone-stimulating peptides in males; prohibited at all times.
- other: Avoid re-exposure after a severe reaction to this peptide or its injection ingredients.
- other: Unexplained low testosterone, high prolactin or absent periods can signal pituitary disease; establish the cause before hormone manipulation.
Interactions
- Testosterone and anabolic androgens: Androgen feedback may blunt the upstream response. KP-10 has not been shown to preserve sperm production during testosterone use.
- Estrogen, progestogens and hormonal contraception: Sex steroids change the LH response. No evidence shows that KP-10 makes contraception fail.
- Other kisspeptins or receptor agonists: Duplicate pathway stimulation has no established combined home dose; prolonged exposure can reduce the hormonal response.
- GnRH drugs and fertility hormones: These act on the same reproductive axis. Research combinations require a set protocol; effects on ovulation and hormone levels may change.
- Insulin and sulfonylureas: No established KP-10 interaction or excess low-blood-sugar rate. Review glucose medicines because metabolic effects remain under study.
Product quality risks
- One analytical study tested one illicit KP-10 vial: no unexpected impurities were found, but it was more degraded than the reference standard.
- One vial cannot establish market-wide purity, correct amount, sterility or endotoxin safety.
- FDA flags aggregates, peptide impurities and immune-response risks in injectable KP-10.
- Health Canada has seized unauthorized KP-10 injections; contamination, wrong ingredients and infection are concerns.
How Kisspeptin-10 works
It switches on kisspeptin receptors on brain cells that release GnRH. GnRH tells the pituitary to release LH and FSH, which signal the testes or ovaries. The response depends on the person’s hormone levels and whether this pathway works.
- KISS1R / GPR54: Activates GnRH-releasing brain cells.
- GnRH → LH and FSH: Changes signals to the testes and ovaries; sex-hormone effects vary.
Half-life: IV plasma disappearance: 3.8 ± 0.3 minutes in men and 4.1 ± 0.4 minutes in women. This is not the duration of hormonal effects.
Gaps: No large trial establishes durable libido, fertility, symptom or testosterone benefits from home KP-10 bolus injections. Long-term safety, immune reactions, cancer risk and cardiovascular outcomes are unknown. No repeated daily, alternate-day or weekly SC bolus maximum has been established; pump rates are not bolus limits. Some registry doses are undisclosed. Per-person titrations and low-dose outcomes shown only in plots were not reconstructed. Community prevalence is qualitative, not a survey. Most hormone claims lack verifiable laboratory records. No validated oral, troche, IM or intranasal KP-10 schedule, or home reconstitution expiry, was found. EU national approvals, UK personal-import rules and Australian KP-10 scheduling remain incompletely verified. Published reports reuse participants; study counts are not independent exposure counts. Registry adverse-event grades were not posted. Display severity bands are symptom triage categories, not measured trial grades.
